TMV-P 외피단백질유전자(CP gene)의 cDNA를 Agrobacterium tumefaciens의 binary Ti plasmid vector를 이용하여 담배(Samsun nn)에 도입하였다. 형질전환된 담배로부터 도입된 CP cDNA는 PCR에 의해서 확인되었다. 선발한 형질전환 담배중, CP gene이 sense방향으로 도입된 형질전환체는 TMV-P를 접종하였을 때 병징발현을 현저하게 억제하였다. 그러나 CP gene이 antisense 방향으로 도입된 형질전환체에서는 비형질전환 식물과 마찬가지로 바이러스의 접종에 따른 병징발현을 억제하지 못하였다. Sense CP gene이 도입된 형질전환체에 증식된 TMV-P의 농도는 비형질전환체에 증식된 바이러스 농도의 1/14이었다. 이들 형질전환체의 자식종자를 발아시켰을 때 kanamycin 내성이 인정되었다.
The cDNAs of tobacco mosaic virus-pepper strain (TMV-P) coat protein (CP) genes were introduced into tobacco plants (Nicotiana tabacum cv, Samsun nn) using a binary Ti plasmid vector of Agrobacterium tumefaciens. These cDNAs introduced into tobacco plants were detected by polymerase chain reaction. Symptom development was distinctly suppressed in the transgenic plant introduced by sense CP cDNA when the plant was inoculated with TMV-P, while in transgenic tobacco plants of antisense CP gene, symptom development was not suppressed as in non-transgenic plants. TMV-P concentration in the sense CP transgenic tobacco plant was decreased to 1/14 of the concentration in non-transgenic plants. Expression of the kanamycin resistance gene of these transgenic plants could be detected in the progeny.
