Short-chain fatty acids (SCFAs), the major end products of fermentation of dietary fibers by gut microbiota, change their levels in a biological system reflect the progress of various diseases. A convenient and sensitive detection method is needed to profile SCFAs in limited and complex biological samples. However, determination of SCFAs in biological samples is limited due to their easy loss during sample workup, high volatility and hydrophilicity, and the coexistence of interference. In this study, we developed a feasible method for the analysis of SCFAs in human plasma by GC-MS combined with flash heater derivatization on SPME fiber. Extraction of SCFAs from plasma sample was successfully performed by SPME method.
